Abstract

This chapter investigated about the Estimation of Beneficial and Harmful Microorganisms of soil.   A research work was conducted at Wild Life Institute of India, Dehradun. Collected samples n-16, where 9 samples excluded from the study due to colored DNA. The 9 sample belong from chemical treat soil sample 7 soil samples (Organic treated one) selected against the Evaluation of Soil Nematodes i.e., Plectus parietinus, Plectus parvus, Caenorhabditis elegans. i.e.,   for the quantity analysis we used Cubimetere (Invitrogen) and all DNA were found to be ranged in 50ng/l to100 ng/µl. the readings of all specimen DNA yield in given in Table 1. The BLAST analysis of selected sample cleared that the all query samples matched with three different species of nematodes. The sample 1 & Sample 4 matched with Plectus parietinus, sample 3, 6 & 7 matched with Plectus parvus, sample 2 & 5 matched with Caenorhabditis elegans. From all the collected Organic and Inorganic Samples, the Nematodes were found from only the samples i.e., collected from organic fields.
