An Overview on Non-steroidal Anti Inflammatory Drugs and Prodrug Concept

The aim of present study is to review on those factors affecting solubility, stability and bioavailability of non steroidal anti inflammatory drugs (NSAID) for possible enhancement of therapeutic index (anti-inflammatory activity) with lesser side effects. That prodrug compound on administration gets proper chemical biotransformation or enzymatic conversion (metabolized) within our body and then converted into pharmacologically active drug. This active moiety provides us better therapeutic index or pharmacological actions in our body in comparison with that of parent drugs. On other hand, these prodrug forms of drugs are able to reduce the adverse effects by masking the carboxylic acid function and other factors positively. On other hand this brings few changes into the pharmacokinetic profiles of drugs that provide us sustained release effects with reduced adverse effects in comparison to that of parent drugs itself. Thus the present article highlights on those parameters for possible utilization of newer prodrugs concept for the purpose of eliminating those limitations of NSAID drugs and moving toward better outcomes and therapeutic index values.
